AGP - (Acclerated Graphics Port) is the physical connector to a Parallel Bus that offeres a direct connection between a computer system's graphics card and the system's chipset.
Parallel Bus: a link between two pieces of hardware in which every line (bit) transfers its data synchronously in conjunction with a clock signal, so an 8bit bus for example transfers 1Byte with each clock signal.
AGP has undergone 3 revisions since its relase in 1997:
AGP 1.0 aka. AGP 1x/2x (66/133MHz @ 3.3v) 1x mode had a data transfer rate of 264MB/sec. 2x mode had a data transfer rate of 528MB/sec.
AGP 2.0 aka. AGP 4x (266MHz @1.5v) 4x mode has a data transfer rate of ~1GB/sec.
AGP 3.0 aka AGP 8x (533MHz @1.5v) 8x mode has a data transfer rate of ~2.1GB/sec.
AGP 1.0 is now considered legacy and is no longer supported by hardware manufacturers. The clock frequencies are mulitplied and operate on the same principal as Intel's "quad pumped" front side bus.
Note: Just so people know since I've seen it all over on this site. There was never a 3x, 5x, 6x, or 7x AGP and there will never be a 9x, 10x, 11x, 12x, 13x, 14x and so on AGP standard. IF they were going to continue it would be 16x. (theres my rant for the day)
PCI-Express (Peripheral Component Interconnect Express) is a connection to a Serial Bus which allows for simultaneous transfers of data to and from the device by the system chipset through the use of Lanes.
Here's how it works. Serial Buses are not really busses in the conventionl way of thinking they are only 1 bit wide. How then do you transfer more than 1 bit of data over a serial bus? Simple, transfer it one bit at a time. Lets say for example you need to transfer 1Byte of data over a serial bus, first the data has to be broken up into 1 bit chunks transfered one bit at a time and put into a buffer at the other end until all 8 bits are there where they are put back together into a Byte.
So how is this faster than a Parallel Bus that transfers all 8 bits in one clock cycle? It's faster than a parallel bus because the clock frequency can be ran much higher on a serial bus than on a parallel bus, because the bits dont have to arrive in synch. On a computer the frequency's are so high that one wire being slightly longer than another on a bus can make the data get there at different times corrupting the information being passed, this is not an issue with serial busses.
PCI-Express uses serial connections in the form of "lanes" each lane consists of 2 signals, a send and a receive. PCI-Express slots are desginated by how many lanes they are wide so a x1 (pronounced "by one" is one lane wide and a x16 is 16 lanes wide.
How fast is it? Each lane can transfer roughly 250MBps in each direction per second. so a x1 slot has a max transfer rate of 250MBps in each direction or 500MBps total and a x16 slot has max transfer rates of 4GBps and 8GBps respectively.
